چکیده

Pathogenic microorganisms possess antioxidant defense mechanisms for protection from reactive oxygen metabolites such as hydrogen peroxide (H202), which are generated during the respiratory burst of phagocytic cells. These defense mechanisms include enzymes such as catalase, which detoxify reactive oxygen species, and DNA repair systems which repair damage resulting from oxidative stress. To determine the relative importance of these two potentially protective defense mechanisms against oxidative stress encountered by Salmonella during infection of the host, a Salmonella typhimurium double mutant unable to produce either the HPI or HPII catalase was constructed, and compared with an isogenic recA mutant deficient in DNA repair. The recA mutant was hypersusceptible to H202 at low cell densities in vitro, while the catalase mutant was more susceptible to high H202 concentrations at high cell densities. The catalase mutant was found to be resistant to macrophages and retained full murine virulence, in contrast to the recA mutant which previously was shown to be macrophagesensitive and attenuated in mice. These observations suggest that Salmonella is subjected to low concentrations of H202 while at relatively low cell density during infection, conditions requiring an intact DNA repair system but not functional catalase activity. Nucleotide and Amino Acid Changes in HN, F and SH genes of an Iranian Mumps Virus; RS-12, Following Attenuation to Vaccine Strain

Background and Aims: Wild-type RS-12 strain of mumps virus has been isolated from an Iranian patient and has been attenuated after several serial passages. This study was designed to determine nucleotide and amino acid substitutions in the HN, F and SH genes during attenuation of the wild-type virus. Materials and Methods: Required viral samples prepared at Razi Vaccine and Serum Institute. Vi...

Sequence and Phylogenetic Analysis of Wild Type Rubella virus isolated in Iran

Background and Aims: Rubella virus is a human pathogen that causes congenital rubella syndrome (CRS) when infection occurs during early pregnancy. Vaccination programs have been remarkably successful in controlling natural rubella infection and CRS. Moreover, ongoing surveillance for all cases of rubella and CRS is a vital component of a prevention program. Although the WHO recommends the use o...

Role of Fimbriae, Flagella and Cellulose on the Attachment of Salmonella Typhimurium ATCC 14028 to Plant Cell Wall Models

Cases of foodborne disease caused by Salmonella are frequently associated with the consumption of minimally processed produce. Bacterial cell surface components are known to be important for the attachment of bacterial pathogens to fresh produce. The role of these extracellular structures in Salmonella attachment to plant cell walls has not been investigated in detail. We investigated the role ...

Impact of the Listeria monocytogenes protein InlC on infection in mice.

The bacterial pathogen Listeria monocytogenes causes serious food-borne illnesses in pregnant women and the immunocompromised. L. monocytogenes promotes its internalization into host epithelial cells and then uses an F-actin-dependent motility process to spread from infected cells to surrounding healthy cells. In cultured enterocytes, efficient spread of L. monocytogenes requires the secreted b...
